The MessageBOOSTER™ cDNA Synthesis Kit for qPCR and the MessageBOOSTER™ Whole-Transcriptome cDNA Synthesis Kit for qPCR enable the user to perform hundreds of sensitive qPCR amplifications from very small populations of cells, even from as little as one cell.
Both kits amplify the mRNA contained in a small total RNA sample and then convert the amplified RNA to cDNA that is ready for PCR or qPCR.
The MessageBOOSTER cDNA Synthesis Kit for qPCR enables the user to perform hundreds of sensitive qRT-PCR amplifications from very small amounts of total RNA… even total RNA from as little as one cell. The kit amplifies the poly(A) RNA in a total RNA sample and then converts the amplified RNA to cDNA that is ready for PCR or qPCR. The MessageBOOSTER cDNA Synthesis Kit for qPCR uses oligo(dT) to prime cDNA synthesis. Therefore, this kit is best used with intact total RNA preparations.

Figure 1. A MessageBOOSTER™ reaction uses a linear RNA amplification process to amplify the RNA from a small population of cells, then converts the amplified RNA to cDNA that is ready for PCR. A reaction can be completed in one day. |
| Amount of Total RNA in a MessageBOOSTER™ Reaction | Number of Real-Time qPCR Reactions That Can Be Performed | |
|---|---|---|
| Low- to Medium-Abundance Transcripts* | Medium- to High-Abundance Transcripts* | |
| 10 pg (~1 cell) | ≥10 | ≥100 |
| 10 pg (~10 cell) | ≥100 | ≥1000 |
| 500 pg (~50 cell) | ≥500-1000 | ≥5000-10000 |
| *Low-Abundance Transcripts = 1-1,000 copies per cell Medium-Abundance Transcripts = 1,000-10,000 copies per cell High-Abundance Transcripts = 10,000-100,000 copies per cell | ||
Table 1. The number of qPCR amplifications that can be performed using cDNA produced by a MessageBOOSTER™ cDNA Synthesis Kit for qPCR reaction is dependent on the amount of input total RNA and the abundance of the transcript(s) of interest.

Applications of the MessageBOOSTER™ cDNA Synthesis Kit for qPCR
- Generate large amounts of cDNA from very small samples of intact total RNA for hundreds of sensitive RT-PCR amplifications.
- Generate large amounts of cDNA from very small samples of intact total RNA for archival purposes.
Benefits of the MessageBOOSTER™ cDNA Synthesis Kit for qPCR
- High Yield. Get hundreds of sensitive qPCR amplifications from as little as one cell.
- Sensitive RT-PCR. Even low-abundance transcripts from total RNA of one cell are readily detected (CT values <35 cycles).
- Less Effort. Collect your small RNA samples less often.
- Time Savings. Reactions can be performed directly from whole-cell lysates without the need for purifying RNA.
- Fast Reactions. Perform a MessageBOOSTER cDNA Synthesis Kit reaction in 1 day.
- High Fidelity. The linear RNA amplification and cDNA synthesis processes preserve the gene expression profile
