To set up PCR simply add equal volumes of DNA, primers, fluorescent dye/probe and run the cycle profile. An initial step of 5 minutes at 95°C is needed to denature the antiTaq polymerase. Since the Taq polymerase is completely inactive at room temperature it is not necessary to set the PCR samples up on ice
