Includes MasterAmp™ PCR Enhancer (with betaine) for improved PCR

MasterAmp™ Taq DNA Polymerase is a thermostable DNA polymerase derived from Thermus aquaticus. Having optimal activity at temperatures above 70°C, it was the first thermostable enzyme used for PCR and remains the predominant enzyme for that purpose. The enzyme has an intrinsic 5'→ 3' structure-dependent exonuclease activity, but lacks 3'→ 5' proofreading exonuclease activity. MasterAmp™ Taq DNA Polymerase is highly purified using a proprietary procedure that virtually eliminates contaminating bacterial DNA. The enzyme is provided with a 10X solution of MasterAmp PCR Enhancer (with betaine). The MasterAmp™ PCR Enhancement Technology* substantially increases both the probability of obtaining the desired amplification product and the reproducibility of PCR. It also greatly improves the consistency of PCR product yields in multiplex PCR. Different concentrations of individual or premixed PCR-tested solutions of deoxyribonucleoside 5'-triphosphates (dNTPs) are also available separately. Alternatively, dNTPs and other components are available in the MasterAmp™ Taq PCR Core Kit.

Unit Definition:

One unit of MasterAmp™ Taq DNA Polymerase converts 10nmoles of dNTPs into acid-insoluble material in 30 minutes at 70°C under standard assay conditions.

Storage Buffer:

50% glycerol containing 50 mM Tris-HCl (pH 7.5), 100mM NaCl, 0.1mM EDTA, 0.5% Tween 20, 0.5% NP-40, and 1mM DTT.

Optimisation of [Mg2+]:

Mg2+ is required for activity of MasterAmp™ Taq DNA Polymerase. We recommend that initial experiments be performed at 1.5 mM MgCl2. The Mg2+ concentration may have to be adjusted for some template/primer pair combinations, or for dNTP concentrations other than 200µM each. Alternatively, we recommend the MasterAmp PCR Optimisation Kit without ammonium sulphate.

Quality Control:

MasterAmp™ Taq DNA Polymerase is tested for the absence of detectable contaminating nonspecific DNA exo- and endonuclease and RNase activities, and is functionally tested in PCR.
