MasterAmp™ PCR Optimisation Kits are designed for those who wish to improve the performance of their current PCR system. These kits are compatible with any thermostable enzymes frequently used in PCR, such as Taq, Pfu, Tth, and Tfl DNA polymerases.
You can significantly improve the performance of PCR systems in one experiment by adding your own enzyme, template and PCR primers into the PreMixes supplied with the kit. This capability is made possible by the newly patented PCR Enhancement Technology.
The MasterAmp™ PCR Optimisation Kit contains a series of 12 MasterAmp™ PCR 2X PreMixes that have been meticulously tested and represent a complete range of PCR conditions frequently encountered. Each MasterAmp™ 2X PreMix contains all components needed for PCR except for the primers, the template, and the PCR enzyme. These components include dNTPs, buffer, varying amounts of MgCl2 and the MasterAmp™ PCR Enhancer (with betaine).*
Choice of Optimisation Kit
There are two types of MasterAmp™ PCR Optimisation Kits available, with and without ammonium sulphate. Choosing the appropriate kit is dependent upon the type of thermostable enzyme used in PCR, since ammonium sulphate is important to the optimal enzymatic activity of certain enzymes and detrimental to that of others. Please refer to the following table to select the appropriate kit for your DNA polymerase.

Three Steps to Optimise Your PCR
Step 1
Perform PCR with your template and primers using your own PCR enzyme in the 12 PreMixes supplied in the MasterAmp™ PCR Optimisation Kit.
Step 2
Select the optimal MasterAmp™ PreMix: At least one of the 12 PreMixes will probably amplify your template/ primer pair.
Step 3
Continue to use the optimal MasterAmp™ PCR PreMix: Subsequent amplifications of the tested enzyme/template/primer pair will work consistently using the PreMix chosen in Step 2.
