The products containing a thermostable DNA polymerase are accompanied by a limited license to use it in the Polymerase Chain Reaction (PCR) and RT-PCR for life science research in conjunction with a thermal cycler whose use in the automated performance of the PCR process is covered by the up-front license fee, either by payment to Perkin-Elmer or as purchased, i.e., an authorised thermal cycler.

The MasterAmp™ Extra-Long PCR Kit is designed for accurately amplifying DNA sequences up to about 40kb. The MasterAmp™ Extra-Long DNA Enzyme Mix is an enzyme mixture, combining MasterAmp™ Taq DNA Polymerase with a 3'Æ5' proofreading enzyme, to achieve PCR fidelity at least three times higher than Taq DNA polymerase alone.
The MasterAmp™ Extra-Long PCR Kit is a complete system that provides everything for successful extra-long PCR except for primers and templates. Included in the kit are MasterAmp™ Extra-Long DNA Polymerase Mix, a set of 9 pre-optimised Extra-Long PCR 2X PreMixes and a control template consisting of a mixture of lambda DNA and primers. The set of 9 Extra-Long PreMixes represents a complete range of extra-long PCR conditions. Tedious optimisation is eliminated through the use of these PreMixes. Each PreMix contains dNTPs, buffer, and varying amounts of MgCl2 and the MasterAmp™ PCR Enhancer (with betaine)*.
Three Steps to Successful Extra-Long PCR
Step 1
Perform PCR with your template and primers using MasterAmp™ Extra-long PCR Enzyme Mix and the 9 PreMixes supplied in the MasterAmp™ Extra-long PCR Kit.
Step 2
Select the optimal MasterAmp™ Extra-Long PCR PreMix.
note: for Premixes A - I, select catalogue number MHF925 with surfix A-I
Step 3
Continue to use the optimal MasterAmp™ Extra-Long PCR PreMix: Subsequent amplifications of the tested template/primer pair will work consistently using the MasterAmp™ Extra-Long DNA Polymerase Mix with the optimal PreMix chosen in Step 2.
