rBst DNA Polymerase Large Fragment is the product of the DNA pol I gene of the thermophilic bacterium Bacillus stearothermophilus (Bst) altered to lack the 5'Æ 3' DNA exonuclease activity. Having optimal DNA polymerase activity at 65°C, the enzyme is suitable for high-temperature isothermal sequencing of DNA. Like the Klenow fragment of E. coli DNA Polymerase I, rBst DNA Polymerase Large Fragment also has strong strand displacement activity, and reverse-transcription activity.
Unit Definition:
One unit of rBst DNA Polymerase Large Fragment results in the incorporation of 10nmoles of dNTPs into acid-insoluble material in 30 minutes at 65°C in 25mM TAPS, pH 9.3 at 25°C, 50mM KCl, 2.0mM MgCl2, 0.2mM of each dNTP, and 10µg activated calf thymus DNA.
Storage Buffer:
50% glycerol solution containing 50mM Tris-HCl, pH 7.5, 100mM NaCl, 0.1mM EDTA, 1mM DTT, and 0.1% Triton® X-100.
10X Reaction Buffer:
200mM Tris-HCl, pH 8.5 and 50mM MgCl2
Quality Control:
rBst DNA Polymerase Large Fragment is free of detectable contaminating DNA exo- and endonuclease and RNase activities.
