FluoroSpin™ Fluorescent Protein Labeling kits are designed for the labeling of proteins, enzymes or antibodies with molecular weights greater than 25 kD. Proteins are labeled using a reactive succinimdyl-ester (NHS) of FAM (Carboxy-Fluorescein), TAMRA (Carboxy -Tetra methylrhodamine) or ROX (Carboxy-X- Rhodamine). A covalent amide linkage forms between the activated dye and protein lysine groups. NHS esters provide superior stability compared to ITC (isothiocyanate) conjugates. The protein-dye conjugates have absorption and excitation maxima that are compatible with the excitation line of an Argon laser.
Up to 1.5 µg of protein can be labeled with the activated dyes. Each kit contains enough reactive dye for up to five labeling reactions. Centri-Sep™ spin columns are provided for removal of unbound dye from labeled protein conjugates. Quick start optimized protocols are provided for the labeling of 100 µg, 1 mg of IgG or up to 1.5 mg protein. Detailed instructions are provided for changing the number of dye labels linked to a protein. Kits are stable for six months in the dark at room temperature (15°C - 20°C).
