The FailSafe™ PCR System is the only PCR system that ensures successful PCR, the first time and every time. It provides accuracy, consistency, and dependability for PCR, regardless of the source or the property of the DNA templates.

This system uses Epicentre®'s FailSafe™ PCR Enzyme Mix, a unique blend of thermostable enzymes that is capable of amplifying the most difficult DNA templates with extremely high sensitivity and high fidelity. Based on side-by-side experiments to determine error rates, the FailSafe™ Enzyme Mix has a fidelity >3 times better than Taq DNA polymerase.

The system includes the FailSafe™ PCR Enzyme Mix, an extensively tested set of 12 FailSafe™ PCR 'PreMixes', representing various PCR conditions. Along with the FailSafe™ Enzyme Mix, these PreMixes contain everything you need for a successful PCR: dNTPs, buffer, and varying amounts of MgCl2 and FailSafe™ PCR Enhancer (with betaine)*.

Incorporating Epicentre®'s patented PCR Enhancement Technology, the FailSafe™ PCR System can accurately amplify:

  • Templates up to about 20kb long - with the minimal number of errors
  • Templates with high GC content (>= 85% G+C)
  • DNA templates present in the reaction at as little as one molecule
  • Multiple templates simultaneously (Multiplex PCR)

Three Steps to FailSafe™ PCR

  1. Perform PCR using FailSafe™ PCR PreMix Selection Kit
  2. Select the optimal FailSaf™ PCR 2X PreMix
  3. Continue to use the selected PreMix for consistent PCR results

Using Epicentre®'s patented PCR Enhancement Technology, the FailSafe™ PCR PreMix Selection Kit has replaced the need for cumbersome adjustment of reaction components to determine the optimal condition for PCR with a specific template and primer pair. The 12 FailSafe™ PCR PreMixes in this kit cover almost all possible PCR conditions that are frequently encountered. Each PreMix represents a unique PCR condition and contains everything one needs for a successful PCR: dNTPs, buffer, and various amounts of MgCl2 and FailSafe PCR Enhancer (with betaine).

The FailSafe™ PCR PreMix Selection Kit: 3 steps to FailSafe™ PCR

Step 1

Perform PCR with your template and primers using FailSafe™ PCR Enzyme Mix and the 12 PreMixes supplied in the FailSafe™ PCR PreMix Selection Kit.

Step 2

Select the optimal FailSafe™ PCR PreMix: At least one of the 12 PreMixes will successfully amplify your template/primer pair.

Step 3

Continue to use the optimal PreMix: Subsequent amplifications of the tested template/ primer pair will work consistently using the FailSafe™ PCR System with the PreMix chosen in Step 2.


The FailSafe PCR System is ideal for any PCR application, such as:   
Multiplex PCR
The ability of the FailSafe PCR System to amplify any template enables more reliable and consistent multiplex PCR (Figure 1).

PCR of "Difficult" Templates
The FailSafe PCR System easily amplifies sequences with 85% GC or other sequences considered difficult with other PCR systems (Figure 2).

High-Sensitivity PCR
With the FailSafe PCR System, you can amplify a sequence present in only 1 molecule of DNA in the sample (Figure 3).

PCR Any Sequence up to 20 Kb from Any Source
The combination of the FailSafe Enzyme Mix, and the FailSafe PCR PreMixes, with our patented FailSafe PCR Enhancer with Betaine, enable robust PCR of sequences and lengths that are not possible with other PCR systems (Figure 4).
