Using Serum in Cell Culture with Confidence

General

The ideal culture medium simulates exact in vivo conditions. In practice this can only be achieved to a limited extent, given the extremely complex nature of the physiological conditions required to cultivate tissues, cells and living organisms.

Serum provides all of the growth factors, vitamins, co-factors, hormones, attachment factors, transport factors, nutrients, trace elements and other factors which limit free radicals, toxins and heavy metals. Serum is a very complex product and only a small percentage of its components have been fully identified. It therefore remains the most effective growth product for cell culture available today.

As a rule of thumb, the more juvenile the animals from which the serum was obtained, the greater the growth-promoting action and the lower the content of antibodies and protein.


Origin

The European Union (EU) will only permit the import of bovine products, including blood and sera, from countries certified by the EU as specific disease free, e.g. Argentina and Brazil.

Sera originating from the USA and Australia are subject to the most restrictive regulations of the USDA (US Department of Agriculture) and are allowed to enter the USA. This is also the case for researchers working on research projects with USA-based researchers.


Further Processing

Our serum is post-treated to make it particularly suitable for specific applications. These treatments eliminate mycoplasmas, viruses and other microorganisms, thus increasing the sera's biological safety.

The use of Foetal Bovine Serum, together with vastly improved serum production processes, has now largely removed heat-inactivate serum. UV light at a wavelength of 254 nm damages nucleic acids, thus killing mycoplasmas and viruses yet not impairing the biological activity of the serum and results in a gain in biological safety greater than that obtained by gamma irradiation. This serum is 100% mycoplasma-free while fully retaining its growth promoting action. As a result, it is ideal for culturing particularly sensitive cell lines or for primary cell culturing.

The sterility and homogeneity of Biontex 100% mycoplasma-free FBS MycoFREE guarantees maximum safety and reliability in cell culture work.

FBS MycoFREE is the ideal serum for all applications where all mycoplasmal contamination must be ruled out and for particularly sensitive cell lines or primary cells.


Storage

Serum is supplied deep-frozen in unbreakable PET bottles on dry ice in a polystyrene foam package. Any serum not used straight away should be stored protected from light at -15°C to -40°C. Partial thawing of the serum before it is placed in storage should be avoided.

Serum may be stored at < -15°C to -40°C for five years with no appreciable reduction in biological activity. Thawed serum can be stored without any problems for eight weeks at 2°C to 8°C.


Thawing Process

When bottles are taken from the freezer, they should be allowed to acclimatise for approximately 10 minutes at room temperature before thawing in a water bath at 37°C. The serum should also be swirled occasionally in order to ensure that the liquid remains homogeneous.

Thawed serum should not be repeatedly frozen, but instead stored in the refrigerator at 2°C to 8°C. If you only require small quantities of serum at a time, we would recommend freezing small aliquots which you can thaw and used as required.
