DisplaceAce™ DNA Polymerase Holoenzyme is a recombinant strand-displacing DNA polymerase derived from a thermophilic bacterium. It has DNA- and RNA-dependent DNA polymerase activity and 5'&arr;3' exonuclease activity. The optimal temperature for the unit activity assay is approximately 65°C. The enzyme can be inactivated by incubation at 80°C for 20 minutes. Thus, if thermal denaturation of a DNA substrate is intended (>75°C), the enzyme must be added after this step to ensure optimal activity.
Unit Definition: One unit converts 10 nmol of deoxyribonucleoside triphosphates into acid-insoluble material in 30 minutes at 65°C.
Storage: Store only at -20°C in a freezer without a defrost cycle.
Storage Buffer: DisplaceAce DNA Polymerase Holoenzyme is supplied in a 50% glycerol solution containing 50 mM Tris-HCl (pH 7.5), 0.1 M NaCl, 0.1 mM EDTA, 1 mM dithiothreitol, and 0.1% Triton® X-100.
