DNA Polymerase I from E. coli is a DNA-dependent DNA polymerase. The enzyme also contains both 5'→ 3'and 3'→ 5' exonuclease activities. The 5'→ 3' exonuclease activity enables the enzyme to use nicks and gaps in the DNA as starting points for labeling the DNA by nick translation.
Unit Definition:
One unit of DNA Polymerase I, E. coli converts 10 nmoles of dNTPs into acid-insoluble material in 30 minutes at 37°C under standard assay conditions.
Storage Buffer:
50% glycerol containing 50 mM Tris-HCl (pH 7.5), 0.1 M NaCl, 0.1 mM EDTA, 1 mM DTT, and 0.1% Triton® X-100.
Quality Control:
DNA Polymerase I is free of detectable contaminating endo- and exonuclease and RNase activities
