E. coli DNA Polymerase I catalyses three different reactions:
- A 5'Æ3'- polymerase activity that requires a template and adds mononucleotides from deoxyribonucleoside triphosphates to the 3'-hydroxyl terminus of a primer chain.
- A 5'Æ3'- exonuclease activity, active only on duplex DNA.
- A 3'Æ5'- exonuclease primarily active on single-stranded DNA.
Typical preparations are greater than 95% pure, as determined by SDS polyacrylamide gel electrophoresis.
Unit Definition:
One unit is the amount of enzyme required to incorporate 10nm of total deoxyribonucleotide into acid-insoluble material in 30 min at 37°C with DNase I activated DNA as the template/primer.
