The AmpliScribe™ T7 Aminoallyl-RNA Transcription Kit produces high yields of aminoallyl-RNA. Aminoallyl-RNA offers a more efficient and cost-effective method for preparing non-radioactive RNA probes for use in microarrays, in situ hybridization, and blotting experiments.

The AmpliScribe™ T7 High Yield Aminoallyl-RNA In Vitro Transcription Kit enables a more reliable, higher yielding and cost-effective method for producing fluorescently-labeled or biotin-labeled RNA for microarray experiments, in situ hybridization and other applications than methods utilizing direct incorporation of a labeled-NTP.

The kit utilizes EPICENTRE's® high yielding AmpliScribe™ T7-Flash™ in vitro transcription technology to incorporate 5-(3-aminoallyl)-UTP (aminoallyl-UTP) into the RNA transcript during an in vitro transcription reaction. Subsequent conjugation of the aminoallyl-RNA to N-hydroxysuccinimide (NHS) reporter molecule, (not provided in the kit) produces a labeled RNA for use as probe. In vitro transcription reaction conditions have been optimized to consistently generate aminoallyl-RNA that, upon conjugation to the fluorescent-NHS or biotin-NHS, yield high signal intensity.

The AmpliScribe T7 Aminoallyl-RNA Transcription Kit produces very high yields of aminoallyl-RNA even from limiting amounts of DNA template Table 1. The AmpliScribe™ T7 Aminoallyl-RNA Transcription Kit produces very high yields of aminoallyl-RNA even from limiting amounts of DNA template.

Applications

  • prepare non-radioactive probes for microarrys (especially those using Affymetrix's GeneChip® or GE's CodeLink arrays), and in situ hybridizations

Benefits

  • The high yields, cost-effectiveness and availability of inexpensive, conveniently packaged Biotin-X-X-NHS make this kit attractive to those preparing biotin-RNA for use with GeneChip arrays
