Diluent: Anhydrous Acetonitrile
Coupling: Use dmf-dG (10-1029-xx) for non-deuterated deoxyguanosine; No changes needed from standard method recommended by synthesizer manufacturer.
Deprotection: Use dmf-dG or UltraMild monomers. Deprotect with ammonium hydroxide for 17 hours at room temperature or deuterated ammonium hydroxide (ND4OD) for 40 hours at room temperature. Technical Bulletin

Stability in Solution: 1-2 days

Deuterated Nucleosides

Perdeuteration and selective deuteration have been useful approaches for simplification of NMR spectra and for other structural studies of large biomolecules. Driven by the progress in multinuclear multidimensional NMR spectroscopy, deuteration of nucleic acids has especially found wide applications in the NMR studies of these complex molecules in solution. 8-Deutero-2’-deoxyGuanosine phosphoramidite will be of interest to our customers involved in NMR spectroscopy.

Notes

Synthetic oligonucleotides containing 8-d-dG must be cleaved and deprotected with 25% deuterated ammonium hydroxide for 40 hours at room temperature to minimise deuterium exchange.
