In Vitro Toxicology: Mutagenicity kits

The Ames II / Ames MPF Microfluctuation Assays are all modifications of the traditional Ames plate incorporation assay.

The Ames II assay uses the S. typhimurium strains TA98 and TAMix (a mixture of 6 strains, each detecting a specific base-pair substitution), whereas the Ames MPF® kits are available with the strains TA98, TA100, TA1535, TA1537, E.coli WP2 uvrA or E.coli WP2 pKM101.

he Ames MPF 1535 assay comprises the histidine auxotrophic Salmonella tester strain TA1535 for detecting base-pair substitutions and all media required to perform the assay. Kits including additional S9 and Positive Controls are also available.

The Ames MPF 1535 assay comprises the histidine auxotrophic Salmonella tester strain TA1535 for detecting base-pair substitutions and all media required to perform the assay. Kits including additional S9 and Positive Controls are also available. 

TA1535 is similar to TA100 but does not contain the plasmid pKM101, whose mucAB gene products enhance SOS mutagenesis. Although the presence of pKM101 makes TA100 in general more sensitive for mutagen detection, there are chemicals mutagenic in TA1535 but not in TA100.
